¸ñÀû: Common aeroallergens implicated in the development of chronic rhinosinusitis include house dust mite derived pathogens. We analyzed differentially expressed genes in primary cultured nasal epithelial cells after treatment of house dust mite and hypothesized that Insulin like growth factor(IGF)-binding protein secreted from nasal epithelial cells are involved in the differentiation of the nasal polyp derived fibroblasts. ¹æ¹ý:The mRNA sequencing analysis on primary cultured human nasal epithelial cells were performed after stimulation with the house dust mite. The validations of the expression of IGF-binding protein in primary nasal epithelial cells were performed using quantitative-PCR, Western blot and ELISA. The effect of recombinant IGF-binding protein on primary cultured nasal fibroblasts were evaluated using various markers of fibroblast activation markers. The expression of IGF-binding protein in human nasal polyps and nasal lavage fluid were evaluated and statistically analyzed. °á°ú:Among various differentially expressed genes, IGFBP3 was prominently up- regulated in primary nasal epithelial cells stimulated with house dust mite extract. The significant difference in the expression of IGFBP3 was shown between control nasal mucosa and chronic rhinosinusitis nasal polyps. Fibroblast to myofibroblast differentiation markers were elevated in primary cultured nasal fibroblast after treatment with recombinant IGFBP3. Knockdown of IGFBP3 receptors using siRNA showed significant decrease in the markers of fibroblast activation markers. °á·Ð:Our result suggests that the IGF-binding protein secreted from human nasal epithelial cells stimulated with house dust mite leads to nasal fibroblast differentiation and involved in the development of chronic rhinosinusitis.
